    摘要:

    细菌性角膜炎是发病急、进展快、致盲率高的常见感染性眼病,诊疗关键是快速精准识别致病病原体并制定个体化治疗策略。当前病原体谱变化及耐药性给临床诊治带来挑战,传统诊断方法难满足需求,下一代测序技术(NGS)凭借高通量、广覆盖、高灵敏度和快速等优势为细菌性角膜炎病原学诊断提供新选择。本文介绍 NGS 技术核心原理与医学应用现状,重点阐述其在细菌性角膜炎精准诊断、耐药性检测和流行病学监测中的应用进展,探讨未来发展方向与临床转化前景,为其在感染性角膜炎精准诊疗中的应用提供参考。

    Abstract:

    Bacterial keratitis is a common infectious ophthalmopathy characterized by acute onset, rapid progression and high blindness rate. The key to its diagnosis and treatment lies in rapid and accurate identification of pathogenic microorganisms and formulation of individualized treatment strategies. At present, the changes of pathogen spectrum and drug resistance of bacterial keratitis have brought new challenges to clinical diagnosis and treatment, and traditional diagnostic methods are difficult to meet the clinical needs. Next generation sequencing (NGS) technology provides a new choice for etiological diagnosis of bacterial keratitis due to its advantages of high throughput, wide coverage, high sensitivity and rapid response. This review introduces the core principles of NGS technology and its current status in medical applications, focuses on the application progress of this technology in accurate diagnosis, drug resistance detection and epidemiological monitoring of bacterial keratitis, and discusses the future development direction and clinical transformation prospects, so as to provide reference for its application in accurate diagnosis and treatment of infectious keratitis.
